About the compound
Retatrutide is a synthetic, long-acting peptide agonist that binds three separate incretin receptors at once: the GLP-1 receptor, the GIP (glucose-dependent insulinotropic polypeptide) receptor, and the glucagon receptor. That third receptor is what separates it from the previous generation of agonists in the class. Semaglutide targets GLP-1 alone. Tirzepatide targets GLP-1 and GIP together. Retatrutide adds glucagon on top, making it the first triple agonist to enter late stage clinical development.
Structurally it is a 39 amino acid peptide, engineered from a GIP backbone with substitutions and a fatty acid moiety attached via a linker for reversible albumin binding. That albumin binding is what carries the plasma half-life out to approximately 6 days, making weekly dosing the operating assumption in the published trials.
Each receptor pulls in a different direction in the research literature. GLP-1 activation drives glucose dependent insulin secretion, suppresses glucagon in the fed state, delays gastric emptying, and modulates hypothalamic satiety signalling. GIP activation adds a separate insulinotropic route via pancreatic beta cells and appears to influence adipose tissue metabolism. Glucagon receptor activation, counter-intuitive in a compound aimed at metabolic control, adds a thermogenic and lipolytic dimension via hepatic and adipose tissue signalling. In Phase II data published to date, the combined effect on body weight and adiposity has consistently outperformed both single and dual agonists tested in matched cohorts.
Research applications span glycaemic control, body weight and adiposity, appetite regulation, energy expenditure and hepatic lipid handling. Both the 20mg and 40mg pens deliver the same molecule at the same 300 unit graduation, so the only variable across the two is concentration per unit.

Handling and storage
- Store refrigerated at 2 to 8°C. Do not freeze
